Two transcritional start sites were mapped in the upstream regulatory region of glgS. An rpoS mutant did not synthesize the glgSp2 transcript. Two shorter fragments visible for the wild-type and the rpoS mutant might be incompletely synthesized reverse transcripts, but might also represent genuine transcripcional start sites indicative of a third and fourth promoters. If so, these promoters would be dependent on cAMP, since these transcripts are not found in the cya strain. glgSp2 is inactive in a rpoS background. Although transcription from glgSp2 requires SigmaS, this promoter does not show similarity to the putative consensus derived from the comparison of the katE, xthA, and bolA promoters. This indicates that either this latter group of promoters or glgSp2 are under the indirect control of rpoS, involving an unknown secondary regulatory protein. |
